CAR-T cell Double Targeting Strategies to Overcome Tumor Antigen Escape and Microenvironment Inhibition
Rationale and Objectives
This project aims to address challenges in autologous CAR-T therapies for relapsed leukemias, including tumor antigen loss, CAR-T exhaustion, and poor-quality products. The goal is to enhance CAR-T therapies using double-targeting strategies and allogeneic HLA-unrestricted gamma-delta T-cells, improving effector cell quality and targeting efficiency.
Thus, the specific objectives of this project are:
- Implementing the design and use of bicistronic CAR-T lentiviral vectors co-expressing two CARs against two tumor antigens or one tumor and one target microenvironment antigen.
- Validating the use of bicistronic CARs in pre-clinical models of T-ALL.
- Upgrading the use of conventional autologous CAR-T to allogeneic-universal gamma-delta T-cells in those cases the quality of donor T-cells is compromised.

This project may be adapted to the evolving needs of the host laboratory while remaining within hemato-oncology research. The DC will undertake one or more secondments with a total duration of at least three months and up to one year. These secondments may take place within or outside the DN, preferably in international and intersectoral settings.
